Detection of marine Vibrio species from environmental samples with a nested PCR technique
نویسندگان
چکیده
Bacteria of the genus Vibrio are indigenous for the marine environment and temporarily in abundances in the ocean, causing infections to humans and commercially important species of crustaceans, bivalves and fish. These bacteria can therefore cause severe complications for human health and large economic losses to aquaculture. A polymerase chain reaction (PCR) technique was used to detect presence of four different Vibrio species from two different seasons in marine environmental water samples, collected in the south west coast of India. Two species, V. alginolyticus and V. harveyi, were detected in both postand pre-monsoon seasons. However, no difference in presence between the seasons could be statistically determined. V. cholerae was not detected in any of the samples and the results for V. vulnificus were inconclusive due to failure to optimize the PCR technique for this species. The qualitative experiment set up was found to be too weak to detect difference in Vibrio species’ presence between the two seasons. However, using nested PCR to detect pathogenic bacteria from environmental samples is an important tool for risk assessments in the food industry.

Molecular Detection of Vibrio spp. in Fish and Shrimp from the Persian Gulf
Vibrio species are the major seafood-borne bacteria that are frequently associated with the consumption of contaminated sea food. A total of 113 samples including 58 fish and 55 shrimps were studied for the possible contamination with Vibrio species. A biochemical protocol was applied for the identification of the Vibrio isolates and Polymerase Chain Reaction (PCR) was carried out to confirm th...
متن کاملDetection of marine pathogenic bacterial Vibrio species by multiplex polymerase chain reaction (PCR).
Polymerase chain reactions (PCR) were performed to accomplish quick and accurate detection of Vibrio species. The primers prepared with 16S-23S rDNA intergenic spacer (IGS) region exhibited an excellent species-specificity for Vibrio sp and detected Vibrio sp more successfully than the conventional culture method. Multiplex PCR was also fruitful not only forthe identification of the 5 standard ...

Background and Objectives: Legionella are gram-negative bacteria widely dispersed in natural and man-made water sources. Some Legionella species are pathogenic and could cause respiratory infections. Cultivation technique is the conventional method for the detection of Legionella spp. in aquatic samples. However, the method has low sensitivity and require prolonged incubation period. Theref...

متن کاملDetection of the phytoplasmal agent of pear decline in Iran, Isfahan province, using nested-PCR
Pear decline is a very important phytoplasma disease that causes considerable quantitative and qualitative losses to this fruit crop. Due to economical importance of pear in Isfahan province, Iran, and the difficulty to determine the occurrence of the disease simply based on symptoms in orchards, a detection method for the phytoplasma causing disease in pears in the region was developed. Since ...
